The COVID-19 ImmunoRank Neutralization MICRO-ELISA is a test for semi-quantitative detection of SARS-CoV-2 neutralizing antibodies in plasma or serum. This assay is intended for detection of circulating SARS-CoV-2 neutralizing antibodies of all Ig classes and to identify individuals who have mounted an adaptive immune response to SARS-CoV-2 indicating recent or prior infection. The assay is designed to detect all Ig antibodies in human plasma or serum that bind to the SARS-CoV-2 receptor binding domain (RBD) and are capable of blocking the binding of the RBD to angiotensin-converting enzyme 2 (ACE2), the viral entry receptor on the surface of target cells.

What are neutralizing antibodies ?

Neutralizing antibodies targeting the SARS-CoV-2 spike protein, specifically the RBD, prevent cell entry by disrupting the interaction between the RBD and angiotensin-converting enzyme 2 (ACE2).

What is the principle of the kit ?

ELISA well A shows antibodies bound to SARS-CoV-2 RBD, preventing binding to ACE2, indicating no color. ELISA well B shows SARS-CoV-2 RBD bound to immobilized ACE2, indicating a blue color.

The ImmunoRankTM surrogate neutralization MICRO-ELISA compares to a live virus neutralization test, such as the Plaque Reduction Neutralization Test (PRNT) and Focus Reduction Neutralization Test (FRNT), but is less laborious, takes only 80 minutes to complete, and does not require a BSL3 laboratory. Current tests for measuring the presence of neutralizing antibodies are not amenable to high-throughput screening and are not widely available in clinical laboratories.
